Two change controls, from the QMS to a go-live date
Northwind Bio makes Veltrimab, a monoclonal antibody registered in 21 markets as a 100 mg vial, a 150 mg prefilled syringe and a 300 mg pen. Priya Raman, its regulatory CMC lead, has a new drug-substance line to sign off — and a new filter supplier has just arrived from the QMS. Here is her morning, screen by screen, in the working solution.
- 01Morning
The global approval runway for Line B at Leiden
Priya Raman · Regulatory CMC leadCC-2026-0417 adds drug-substance Line B at Plant 3 — Leiden, a duplicate of Line A. The runway shows one bar per market from submission to expected approval: 36 registrations touched in 19 markets, 19 filings, the US CBE-30 and the EU and UK Type IB first, in Jan ’27. South Africa's Type II amendment is the longest runway at 30 months. South Korea and Mexico register a different site, so they are not affected.
Go-live suggestion: “Apr 2028 · Q2 2028 · 18 of 19 markets · 96.1% of volume — South Africa follows when approved.” The change control had planned Mar 2027, 13 months before the approvals.
- 02The assessment
Every registration, presentation by market
Priya Raman · Regulatory CMC leadThe impact matrix colours each registration by what its country needs: 10 prior approval, 9 notify and wait, 2 not affected with a supply restriction, and 4 that need her call. The trace shows how the agents got there — the change reader in 4 s, the registration finder with 39 registrations in 21 markets, the dossier matcher quoting the registered text and finding 1 dossier out of date.
What needs you: “Türkiye · Type IB variation — Registered detail out of date — correct in the same filing.”
- 03A judgement call
Japan: a partial change approval, at 71% confidence
Rule classifierThe registered text is quoted from the approval matters: “The approval matters for the manufacturing method state the culture step runs in one 12,000 L production line at Leiden.” The PMDA consultation record from March 2023 sits beside it. The classifier proposes a partial change approval — about 12 months, submitted Dec 2026, approved Dec 2027 — and names the alternative it considered.
“Alternative considered: Minor change notification — if PMDA agrees the line count is descriptive, not an approval matter. Ask Kenji Mori to confirm in a pre-consultation.”
- 04The filing plan
When Line B material can ship, and where
Runway plannerTarget submission is Dec 2026, once the Line B conformance batches and comparability report are ready. First approvals arrive in Jan 2027, 90% of volume in Feb 2028 and the last market, South Africa, in Jun 2029. The share-of-volume curve tops out at 96.5% — South Korea and Mexico must never receive the new material.
Dates come from three years of the company's own approval history and its sequencing rules — Saudi Arabia files after the EU decision.
- 05The memo
The regulatory impact assessment, drafted for signature
Assessment writerThe memo follows the company template: the change, the registered elements touched (3.2.S.2.1 and 3.2.S.2.2), and a filing table by market with the rule-book basis and expected approval. It stays a draft while markets are still proposed. Ten markets need prior approval, so Dr. Maya Chen adds a second approval after Priya signs.
“DRAFT · 21 markets still proposed” — only decided markets go into a signed memo.

A new change arrives: a second sterilising-grade filter supplier
Niamh Doyle · Change owner, Plant 5 — CorkCC-2026-0431 qualifies a second supplier of 0.2 µm PES sterilising-grade capsule filters for the final filtration of Veltrimab on filling Lines 1 and 2 at Cork — same pore rating, membrane and filter area. The plant has planned to implement in Apr 2027. Priya presses “Build the runway”: the change reader, registration finder, dossier matcher, rule classifier and runway planner run in turn.
Change reader: “add an alternate supplier of sterilising-grade filters (drug product) · 3.2.P.3.3 · 3.2.P.3.5.”
- 07About two minutes later
Twenty-one bars, and China's is the longest
Runway planner39 registrations in 21 markets, 21 filings: South Africa and Saudi Arabia implement and tell, most markets notify and wait, and South Korea, Singapore and China need prior approval. China's supplementary application runs to Feb 2028. The go-live line slides from the planned April to September.
“Sep 2027 · Q3 2027 · 20 of 21 markets · 95.0% of volume.” Planned in the change control: Apr 2027 — 5 months before the approvals.

China, with the registered text and the rule cited
Rule classifierThe NMPA registration file registers the sterile filtration filter supplier in 3.2.P.3.3 and 3.2.P.3.5. China's rule book, v10 of Jun 2026, is quoted beside it. The proposal is a supplementary application (major), 80% confidence: about 14 months, submitted Dec 2026, approved Feb 2028. Li Wen is the affiliate.
Rule book · China: “A change of sterile filtration materials for a biological product is a major change: file a supplementary application with filter validation, extractables data and three batches; median review about 14 months.”
- 09Decided
September is set; China keeps the current filter
Priya Raman · Regulatory CMC leadPriya chooses the split the runway planner suggested and sets Sep 2027 as the go-live date. The date is written back to the change control and supply planning is told. China keeps the current filter until NMPA approves in Feb 2028 — one date everywhere would have meant waiting until then.
“CC-2026-0431: go-live Sep 2027 written to the change control · supply planning told.”

One filing task per market
The affiliatesWhen an assessment is approved, a task opens for each market with the category, the rule and the documents needed — owned by the affiliate, with a submit-by month and an expected approval. Kenji Mori, Li Wen, Ana Souza, Hanna Berg and Lena Ortiz work them from one tracker: 29 to prepare, 18 submitted and waiting.
